In vivo and in vitro effect of FK 506 on rat Leydig cell function.

چکیده
FK 506 is a macrolide antibiotic with powerful immunosuppressive activities. It is known to prevent allograft rejection in rats, dogs, monkeys, baboons, and humans. 1.2 In vitro studies indicate that, like cyclosporine (Cy A), FK 506 acts specifically by inhibiting the transcription of a limited set of early T-cell activation genes, including those encoding interleukin-2 (IL-2), IL-3, IL-4, and interferon-gamma (IFN-y).3 There are reports indicating that CyA impairs testicular steroidogenesis and spermatogenesis in rats.4 •5 The immunosuppressive effect of FK 506 is many times more potent than that of Cy A and to date there are no published studies concerning toxicity of FK 506 on testicular functions. The present study was undertaken to assess the in vivo and in vitro effects of FK 506 on rat Leydig cell function.

Intra-CA1 administration of FK-506 (tacrolimus) in rat impairs learning and memory in an inhibitory avoidance paradigm
Objective(s): Calcineurin (CN) is a main phosphatase and a critical regulator of cellular pathways for learning, memory, and plasticity. The FK-506 (tacrolimus),a phosphatase inhibitor, is a fungal-derived agent and a common immune suppressant extensively used for tissue transplantation. To further clarify the role of CN in different stages oflearning and memory the main aim of this study was t...
متن کاملEffect of FK 506 on glucose metabolism and insulin secretion in normal rats.
FK 506 is a powerful immunosuppressant currently being investigated in clinical transplantation. I In vivo administration of this drug resulted in the prolongation of skin, liver, kidney, and heart allografts in large and small animal models.2•3 The immunosuppressive effect has also been shown recently to be effective in the prevention of development of diabetes in BB rats.4 In baboons. treatme...
متن کاملFK-506, a novel immunosuppressant isolated from a Streptomyces. II. Immunosuppressive effect of FK-506 in vitro.
The immuno-pharmacological profile of a novel immunosuppressive agent, FK-506 produced by a streptomycete, is presented here. We proceeded to test the effect of the agent on various in vitro immune systems. It showed that mixed lymphocyte reaction, cytotoxic T cell generation, the production of T cell-derived soluble mediators such as interleukin 2 (IL-2), interleukin 3 and gamma-interferon and...
متن کاملP-4: Adverse Effects Associated with PersistentStimulation of Leydig Cells with hCG In vitro
Background: The detrimental effects of persistent stimulation with hCG were investigated in rat Leydig cells in vitro. Materials and Methods: Oxidative stress parameters such as lipid peroxidation, Reactive oxygen species(ROS), Total antioxidant capcity (TAC) and activities of antioxidant enzymes were assessed in the treated cells. Apoptosis amongest the trearted cells were detected by in situ ...
متن کاملIn-vitro interaction of a novel immunosuppressant, FK 506, and antacids.
The effect of selected antacids on the amount of FK 506 in solution in simulated gastric juice has been studied. FK 506 (2.5 mg) was incubated in 100 mL simulated gastric fluid (SGF) with the equivalent of 500 mg of various antacids. The addition of Mylanta and Tums resulted in 14 and 30% loss of FK 506, respectively, in 24 h; 98% loss was observed in 12 h in the presence of Mag-Ox; 100% loss w...
